nanoCUT&Tag chromatin profiling,
14-18 July 2025, SU

The aims of this workshop were to educate SCENTINEL members in constructing libraries and analyzing nanoCUT&Tag datasets, based on the protocol developed by the Bartocovic lab at Karolinska Institute Stockholm
dx.doi.org/10.17504/protocols.io.8epv59o8dg1b/v2 with adjustments to Drosophila tissues developed by the Mannervik lab.
a) Bioinformatics workshop: introduction and analysis of nanoCT datasets, code sharing to SCENTINEL partners
b) Wet lab workshop: construction of nanoCT libraries

Bioinformatic workshop (14-15 July)

Single cell epigenomics workshop (Sergei Pirogov, SU)
nanoCT data pre-processing (Artem Ilin, SU)
Hands on analysis of nano scCUT&Tag data (Aleksander Purik, SU)

Wet lab workshop, constructing nanoCT libraries (16-18 July)

We tested this newly developed protocol with different Drosophila material
a) embryos
b) 3rd instar larval brains
c) tumor material allografts, propagated in host flies

We followed the nuclei extraction protocol developed by Artem Ilin (SU) https://www.protocols.io/view/nuclei-extraction-from-drosophila-embryos-x54v9o4m4v3e/v1 and the nancoCT protocol developed by Bartocovic lab dx.doi.org/10.17504/protocols.io.8epv59o8dg1b/v2
